Il fut le deuxième fils du prince de la maison d'Ascanie Auguste d'Anhalt-Plötzkau et de sa femme Sibylle, la fille du comte Jean-Georges Ier de Solms-Laubach.
1653, à la mort de son père, il gouverna avec son frère Emmanuel pendant douze ans.
1665, il épousa Sophie Eleonore, la fille du comte Henri Volrad de Stolberg-Wernigerode et de sa seconde épouse Margarethe.
1669, avec son frère, ils abandonnèrent la principauté d'Anhalt-Plötzkau pour la principauté d'Anhalt-Köthen.
Il est inhumé à (voir en anglais, église saint-Jacob) de Köthen.
